Will the iPhone 4 play through a car stereo using only the USB interface, or is it necessary to also use a cord from the headphone jack to the stereo input?

I don't think it will play from the usb, I used the audio jack to play music using the car system.
It depends on the stereo ... if it was designed to support iPod/ iPhone (and is up to date to support current models) then yes just connect to USB with a standard dock cable. If the stereo does not specifically state that it supports iPod or iPhone, then it likely won't work.
It depends entirely on the system you have installed in your car.
We have Kia's that have a USB interface that can be used to play from the iPhone. In our case, it requires a special adapter cable from Kia to work.
Personally, I usually just stick with Bluetooth.
