Are you plugging the iPod into a high powered USB 2.0 port on the back of the PC and not into a USB hub?
Do you have another USB cable for your Shuffle that you could test it out with to rule ou the possibility of a faulty cable?
If you have any other USB devices plugged in, remove them all. From there, you should reboot your PC and reconnect your Shuffle after opening iTunes.
Sometimes, a hard reset of the iPod helps as well.
