I can see that to one iPhone you may have been having issues if it is not maintaining a connection.

In System Preference > Network > Advanced button > DNS tab add at least one Public DNS server
Google run at least two with 8.8.4.4 and 8.8.8.8

With the Finder as the "Front" app in the Menu Bar use the Go Menu.
In here with the menu bar in drop down mode, hold the ALT key to display the Library option.
In the Library open the Preferences folder.
Find com.apple.imservice.iMessage.plist and drag it to the Trash and then restart Messages.

Hopefully 10.8.1 fixes it. If you have any other suggestions, I'd appreciate all the help.

I've been following this discussion because I have the same issue. I have tried removing the plist file as advised earlier also with and without one of the public Google DNS addresses you recommended.
I have just tried again and the message failed to be sent the first time. If I click on the red exclamation point and select Try Again it will be sent successfully. I would like to 'fix' it if possible.

I have removed my Google DNS server and "Sent" an iMessage to my phone.
No problems were seen.
I also tried to the iPhone's number as well as the Apple ID.
Same result.
I would also try a PRAM reset.
Shut down the computer.
Restart it holding down Apple/CMD ( or ⌘)+ALT( ⌥)+P+R Keys until you have heard three Start Up Bongs.

I found that I cannot reach my website or the Service provider for it if I remove the Google servers.
This is most bizarre as I have been with them longer than I have been using 8.8.8.8 and 8.8.4.4
But as soon as I put them back it I could get there.
I may receive some form of compensation, financial or otherwise, from my recommendation or link

The issue seems to have re-surfaced. I have no problems sending to my own iPhone but sending a message to a friend's iPhone just now resulted in the red exclamation point. It did send successfully the second time.
If you have any other thoughts I would greatly appreciate hearing them. Perhaps the release of IOS 6 will solve this.
