I had the same problem both at home and work. Changed my DNS server to 8.8.8.8 and the problem went away.

Since home is Comcast and work is corporate, I'm not sure why they both had the issue (the corporate proxy servers are on the east coast, I'm in the mid-west) but changing DNS fixed it for me. It then found and downloaded the update for iTunes to 10.7.

Could be a temporary issue with both Comcast and our corporate DNS servers I guess??

Also - after applying the update, and then changing the DNS back to the default, checkign for updates works fine, no more error.
