If you load songs manually, by dragging songs to the iPod in iTunes, this is what you do.  Select the iPod in the iTunes sidebar, under DEVICES.  Click the little triangle next to the iPod, so that it points down.  This "drops down" a list of media types (indented under the iPod).  Select Music there.  Over to the right, you should see a list of songs that are currently on the iPod.  Select the song or songs you want to delete on the list, and hit the Delete key.  NOTE:  You can select multiple songs at the same time.

If you use automatic syncing to load your iPod, the sync is based on how you set up the Music tab.  Select the iPod in the sidebar, and over to the right, go to the Music tab.  For automatic syncing, this is where you tell iTunes how to sync your iTunes music library to your iPod, based on playlists, albums, artists, or genres.  For example, you can create a playlist (or more than one playlist) in iTunes with only the songs you want on the iPod.  Then, set the Music tab to sync that playlist.  Going forward, when you want to update the songs that are on the iPod, just update that playlist.  The next time you connect the iPod (or click the Sync button), iTunes will sync that change to the iPod automatically.  NOTE:  If you have songs on the iPod that are not in your iTunes library, using automatic syncing will delete those songs.
