I am also experiencing the same issue as of September 12, 2012. Anytime I attempt to check for software updates, I end up with the error message:

An error has occurred

The operation couldn’t be completed.

(NSURLErrorDomain error - 1100.)

I am not sure if there is something we as users can do on our end, or if it’s a technical issue on Apple’s end with their servers.

LOTS of people are receiving this error (including myself). It probably has nothing to do with your computer at all but the fact that Apple's servers probably weren't prepared for the massive numbers of attemped updates. DNS fixes have been posted, but these don't work for me. Unfortunately, probably the best solution is either to wait or, if you're looking for the update to iTunes, go to: http://support.apple.com/downloads/ where you can find the latest software.
