When it is released later today - if you are running iOS 5 - go to Settings>General>Software Update and you can update OTA via WiFi.
If you are not running iOS 5, you need to connect the iPad to your computer and update via iTunes.

The update is not automatic. You must initiate and install the update like I described above - in the iPad settings.

The way that you decide to update is up to you. I plan updating via WiFI myself and not use iTunes. You have the choice. If you decide to update with iTunes, you may want to temporarily disable your firewall and antivirus software until the update completes. But you can update over WiFi if you want to do so.
Remember that millions of people world wide will want to update their devices and the servers may be very busy today.
When iOS 6 is released later today you should be able to update either via Settings > General > Software Update (I haven't seen anything that says you can't update to iOS 6 directly on the iPad) or via your computer's iTunes - but doing it directly on the iPad should just download the parts of the update that are needed (i.e. the bits that need updating), doing it via your computer's iTunes will download the complete iOS so will probably be a lot larger.
Yes you can do it OTA(over the air) or if you don't have wifi, you can use iTunes.
