How to use Mac with 2 hard drives?

I have a MacBook Pro that I have been using with one hard drive for 2 years. I am about to install a second hard drive inside of the computer, and I would like to know how use my laptop with these 2 hard drives. The drives I will use will be...

60 GB SSD (primary)

OS
Applications

750 GB HDD (secondary)

iTunes library
Docs, pictures, music, movies, misc.
Everything else

Is this possible? Do I need to simply drag and drop the folders onto the hard drive that I want them in?

By default the HDD will be excluded from Time Machine backups. Open Time Machine Preferences and remove it from the exclusions list and then it will be backed up along with the SSD.
