Quit iTunes first.
In Computer, open Local Disk C: or whichever drive you have your program files installed on.
If you have a 32-bit Windows, open the "Program files" folder.
If you have a 64-bit Windows, open the "Program Files (x86)" folder.
Open the "iTunes" folder.
Right-click on the iTunes.exe file and select "run as administrator".
Try playing the movie now. Does it play for you without the message?

Let's try running iTunes both as an administrator, and in Safe Mode.
Quit iTunes.
Head back to your iTunes.exe, but this time right-click the itunes.exe and select "Properties". In the Compatibility tab, check "run as administrator" and click OK.
That's automatically start iTunes as an administrator.
For starting iTunes in Safe mode, see the following document:

Any better luck when running as an administrator in safe mode?
