How can I update my iPhone 4 to version 6.0 when my phone will start to back up but half way through it comes up with a message saying an error occured while backing up iPhone (-50) if you continue it will result in loss of contents on iPhone

If I remember correctly, you are asked by the uninstallation routine, whether you want to delete your iTunes library when uninstalling.
