I have a 27 inch iMac and want the icons and text to be larger
I also want this to apply to my other applications such as office
the command+ only increases the active email screen it does not keep it the next time I enter the application and doesn't work on the ribbon acroos the top

Can't be done unless you change the screen resolution.
As Klaus says, changing screen resolution will give a universal effect. I would add, icons can be enlarged with your existing resolution, in the Finder windows in icon view (by adjusting the slider to the right, on 10.6 here) and on the Desktop, (show view options, Icon size).
Click on the Desktop, CMD+J, and view options window pops up. That allows resizing icons and selecting the text size, but won't change the size of text in the menubar.

The resolution change did make the text larger and I cannot seem to get the text any larger than 16 using your method

16 is a lot larger than the default 11 or 12. I have mine set at 11 using the 2560 x 1440 resolution setting, but then I've 20/20 peepers.
