I have two laptops that are networked. Both have their own iTunes files but also have home sharing. Can I sync my iPhone to either laptop or only the one in which i set the phone up on?

You can sync your iPhone only with one computer. It does not have to be the one you set it up on and you can change the computer if you have to.
While syncing your iPhoone with another computer the previous sync will be removed together with libraries, music etc.
You can use iCloud for better results -depending what you want to achieve.
