There's no need for "optimization" in OS X.
If you do a lot of video editing it may be worthwhile. The bet way to optomize is to clone your drive to another disk, erase your disk and then clone back. No fragmentation, clean as a whistle. Best the optmization utilities.
EDIT: I wouldn't do it with a TM drive though.

Because Mac OS handles optimization itself. All I could dig up right now is an archived article...
