Chances are, you already know how to connect the printer to the USB port on the AirPort Express.
What you really may be asking is how to configure things so that you can print this way.
On your Mac....
Open System Preferences (gear icon on the Mac's dock)
Open Print & Scan
Click the + button at the bottom of the printer list
Hopefully, your printer will appear after a scan for printer drivers
If yes, click the name of the printer to highlight it
Click Add at the lower right
Try to print. If you have previously configured the printer to work when connected to your Mac, you actually have two "locations" for the printer now installed, so you must choose the correct "location" when the print dialog box appears.
Using the Print & Scan box, you can also designate the printer connected to the AirPort Express as the "default" printer, so you won't have to choose which printer to use.
If your printer does not appear after a scan for printer drivers, you might try contacting Brother support to see if they possibly have a printer driver that you could download and install to enable the feature that you want.
If you still cannot print, you should understand that not all printers are compatible when connected to the USB port on the AirPort Express, so that may be the case here.

I got the mac to print to the Brother via the AirPort, but the iPad still does not recognize that there is a printer connected to the AirPort. It looks for a AirPrint printer.
You need to install an App like Print Central on the iPad to see if that will allow printing. No guarantees, but it is probably worth a try if you need to print from the iPad.
