iTunes error (-5000) during update of my iPad from iOS 4.3 to iOS 6.0.1

During update of my iPad from iOS 4.3 to the latest version of iOS 6.0.1, iTunes gives an error (-5000) while backing up the iPad and asks "Would you like to continue? Continuing will result in the loss of all contents on this iPad."

Has anyone else seen this? Any suggestions for next step?
