Can the external hard drive be in FAT format to back up the hard disk to and external hard drive and will this erase any other data already on the external disk?

Basically, no; while loose files such as individual photos can be backed up from a Mac to a FAT32 drive, it won't work with all file types and can't be used with either Time Machine or as a bootable clone.
