What are you referring with that you have locked your computer? If this means that you have forgotten the password, see > http://support.apple.com/kb/HT1212
If you have OS X Lion or Mountain Lion > http://discussions.apple.com/docs/DOC-4101
I have changed the setting from "ready only" to "no access" thinking that this would block access from other users to my macbook. Now I have no access to my Air. I didn't realized that I am an everyone! :)
If you do it only for your user, you can try resetting permissions > http://osxdaily.com/2011/11/15/repair-user-permissions-in-mac-os-x-lion/
If this doesn't work, open Disk Utility from Recovery (if you have OS X Lion or Mountain Lion, pressing Command and R keys while your Mac is starting) or the Mac OS X DVD (if you have Snow Leopard or older, pressing C key while your Mac is starting), select Macintosh HD in the sidebar and repair permissions.
If this doesn't work, reinstall Mac OS X
