Depends on the site. When on the mobile version of the site, look for a link to their desktop version. If none is offered, it's not possible with Safari.
There are other browsers in the App Store that allow you to change the browser ID to somehting other than Mobile. iCabMobile is one that allows you to set the ID to any one of many browsers, tricking the visited site into thinking you're using a desktop browser, thus automatically displaying the desktop version of the site.

I actually discovered that I could change the orientation to desktop by rotating the phone from north/south to east/west.
For some unknown reason, I don't have to do this with my iPad.

When I went to the app store, I was unable to locate iCabMobile. Are you sure you typed the name correctly?

Maybe it's not available in your country. I found it in Greek App Store.
Although Safari does not have a setting for desktop mode, Google Chrome (free app) does under menu option "Request Desktop Site".
