I have a @comcast.net email address. Can I use icloud to manage/sync mail across a desktop PC, PC laptop, iPad and iPhone using my current @comcast.net email address?

iCloud provides a @icloud.com address that you can set up in all your devices, but you can't use iCloud to do it with your @comcast.net mail address. In this case, you will have to set up this mail address manually in each computer and device
Another thing to keep in mind is that Comcast email is a POP email account. POP accounts don't keep your email in sync across your devices. Each device will operate independently and actions taken on one device (such as reading and deleting messages) will not be reflected on your other devices. Only IMAP and exchange accounts (such as iCloud email, Gmail, etc.) will do this.
