I have a simple label document in Pages which I print to my label printer. The label is 1.49 x 3.47 inches and needs to print in Landscape, and it needs to print to my Dymo label printer. How do I set up the Page Setup or Print Setup so it defaults to this document size, orientation, and printer, every time, without me having to manually enter those settings every time?

That's what custom Templates are for. Once you get everyting set the way you like it, File > Save as Template.
Do a print with your master document before saving it as a template and Pages is supposed to remember the printer choice as well. Unfortunately, Pages has often been rather forgetful with regard to the printer choice.

Thank you for your reply. However, I could not get Templates to work correctly for me. Following your instructions, I opened my document, opened Page Setup, set the printer, paper size, orientation, and printed a copy to make sure it was right. Then I re-opened Page Setup to verify that the settings were still correct. Then I saved the document as a Template. Then I found my template in Pages, opened the template, and tried to print. The first thing I noticed was that the template did not remember my printer. Then I set the printer, and printed, but I found that the template also did not remember the page size or the document orientation. So, did I miss a step here? Lion 10.7.5 and Mac Mini.
The templates doesn't remeber printer settings but documents do! Save it as a document instead.

Templates definitely should remember page orientation and size.

The templates can't know which printer you want to use. Also when first havng saved a document and then print it Pages asks if you want to save the document. The only thing that has changed is that you have used a defined printer.

Page Orientation and size are Document Properties, and since they affect layout they must be saved with the template. Printer choice is another matter and should not change any aspect of the document.
Since even documents are not reliable at remembering printer choice, even though it appears they do try, I don't know how we could confidently say that templates do or do not try to remember the choice.

I can confidently say that the templates doesn't remember the printer as it never has used one!
Keep in mind that this is a Custom Template made from a normal document prior to being saved in My Templates. To me, it's a minor concern. How big a problem could it be to make a printer selection in the Print Dialog on the occasions when labels are to be printed?
