I have lots of apps that i no longer use or have found better ones to replace them  - how do I delete from the mac they are stored on?

You can delete them from the Apps section of your Mac's iTunes : select Apps on the left-hand sidebar under 'Library' (on iTunes 11 you can enable the sidebar via option-command-S), and then on the right-hand side you can select the apps you no longer want and press the delete button on your keyboard.

You can't currently permanently delete any items from your purchase history, but you can hide them so that they are less visible (e.g. they won't appear in the Purchased tab for re-downloading) : hiding and unhiding. If your iPad is on iOS 6 then you will need to do the hiding via your computer's iTunes, it doesn't currently work directly on iOS 6 devices.
