I have quit aperture and reopened.
Shut-down computer then reopened.
Tested under a new library and a new user and the problem still persists??

Then reinstall Aperture. What version did you update from?
It can be, that the download was incomplete and did not install correctly. Do you have any plug-ins installed, that may not be compatible with Aperture 3.4.5?
BTW, are your original image files raw? Are your Aperture libraries on the system drive or on an external drive?

If Showglow Pix tested from a new user account, then it is unlikely that removing the user preferences will help - that is indicating a system wide problem - broken Aperture application, incompatible plugins, kernel extensions, helper applications, system trouble, hardware trouble.

re-installed software, updated to 3.4.5 and still the brush adjustments don't work and straighten tool don't work.
is it possibly something to do with an incombatiability between the new update and a plug-in?

Which plug-ins are you using? Are they all compatible with Aperture 3.4 and Lion?

I've tested in a new library and it still continues.
The plug-ins by the way all work fine. It's solely any adjustment brushes, the straighten tool, crop tool and patch tool. Any color or exposure adjustments which I add work fine.
It seems any adjustments which affect only a portion of the image or affect the images size dimensions are being prevented from working. Remembering that whenever I select one of these unusable adjustments it is accompanied by the "funk (beep)" apple sound for an error.
Please help. I'm currently having to perform the above adjustments through Photoshop but it's such a time suck!

And do you still get the beeps when working from a different account ?
Check your version of the Nik plug-in you are using. And check your Arial fonts:
