I have the newest ipad and whenever I go onto the safari and click on a streaming video the play button comes up, but has a line through it this means i can not play the video. What do I have to do to make it play the video.

If the site uses Flash based videos then it won't play natively on the iPad. Mobile safari doesn't support flash. You'd need to use a flash based browser, I see Puffin, Skyfire and iSwiffer recommended. But none of them seem to be perfect, so you may want to experiment before you plunk down any money.
You might also want to see if the site you're trying to access has an app.
