What OS are you using? If you are on the Lions (10.7 or 10.8) this is the alternative:

Have you checked in your System Preferences that Spaces shortcuts are enabled?
Sometimes if your using the keyboard for shortcuts (like the Fn keys) they can get thrown off by something like the Caps lock etc.,
Have you tried double clicking on the Spaces program in the Applications/Utiltiies folder (yes it does run!)
It could be the program for it is corrupted or there is some sort of permissions issue.
Please run through this list of fixes and tune your machine up or have a local professional take a look at it.
