If your Mac is running Lion or Mountain Lion: go to System Preferences>iCloud and sign in. Check 'Mail' in the checkbox there.

If your Mac is running Leopard or Snow Leopard you can access email on an existing iCloud account (you cannot set up a new iCloud account with these) by following the procedure detailed here:
