Do I need to activate my iphone 5 from barcelona before I use it in Israel?
I buy a new IPhone 5 in Barcelona store and I don't activate it in the store because I want
to unboxed it in Israel and activate it here do I will have any problem because of this mistake?
If I am unboxed it here and start the activation, It will work with my Israely carrier?

If it's locked to a carrier, activation has to be done where there is local access to that carrier, i.e.in Spain.
If it's unlocked, it can be activated in Israel using a SIM card from an Israeli carrier
and if it locked to spin Movistar carrier

If it is locked to Moviestar in Spain ,it will not work in Israel with a local sim
it will only work roaming from Moviestar at exorbitant rates
If you have purchased take it back it is useless for full time use in Israel
Forget activating it just will not work in Israel

No, you can't activate it with Movistar while in Israel, or use it with and Israeli carrier. You'll have to return to Spain, and activate it with Movistar there. You can then use it in Israel if international roaming is provisioned on your Movistar account, but this is extremely expensive.
An alternative to get Movistar to unlock it for you, if you meet their requirements. You would need to have a Movistar account and have activated the phone with them first.

Who is "she" who told you it's unlocked? The websites with unlocked listing are not reliable. The only source to be trusted is Apple. Call AppleCare (Google to find the number)

All Apple Stores in EU sell unlocked or they will sell with a carrier contract
