OS X 10.8 Mountain Lion
System Preferences > Dictation & Speech > Text to Speech
1. Put a checkmark in the box beside
"Speak selected text when the key is pressed
current key: Option + Esc"
2. Highlight the words and press option+esc keys.
You can change the keys if you want.

Thanks for your reply but I've already tried that and it hasn't worked. When I highlight the words and press command-esc to words are no longer highlighted and it won't read the words. It seems to only happen on the Kindle application. Do you have any other ideas of how I can resolve it.

Press the Reader button in the url bar for easy reading.
Note the sentence "Click and drag............"

The Kindle app has its own options when highlighting text - and Text-to-Speech is not available.
I 'spoke' too soon - highlighting and pressing option-escape does work for me, regardless of the apps 'options'!
