Rebuild the Spotlight index. If you try to search now from the magnifying-glass icon in the top right corner of the display, a pulsing dot will appear in the icon. When the dot disappears, the indexing is complete.
Will this also free up the space on my hard drive?
Ultimately nothing is going to clear up space on your mac but getting an external HD and offloading large files for storage that dont need to be on your Mac.
Storage is cheap. $65 for a 1TB drive

No, but if you're getting inaccurate Spotlight results it may solve that problem.
First, I appreciate everyone's help. Still having issues though. I had already purchased a hard drive and dumped my video files. When looking at my Macintosh HD it shows 77 GB labeled as movies. When I look at my files though I only see about 32 MB (with an M). How can this be? When I try to upload more videos into iPhoto it stops short and says there isn't enough space.

If you're backing up with Time Machine, turn it OFF in its preference pane, then turn it back ON. Try again.
