Genius not working

I'm having a problem with genius on my itunes, I'm using the latest version and I regularly update but for some reason genius will not work with a single song out of my 50gb collection, most of it is from itunes and all is listed on the itunes store. Any ideas, (I have an ipod touch 5th gen and the same problem on there as well).
