What version of the iOS are you using and where are you blocking the number?
If you are blocking the number through your carrier it may take some time for the unblock to happen.
If you are using the Block fewture in iOS 7 you should see the unblock happen right away.
I am using OS7 and the block appears to be removed immediately as displayed on the phone. However, the effective un-block on the number does not occur immediately. This has been tested for time both for the block and the un-block. The block is immediate but the un-block does not occur for some time (yet to be determined).

Reset the iPhone.
I found that the block is immediate, but the un-block takes an undetermined amount of time.
Happens immediately.
Not according to Apple.
That's interesting. As soon as I unblock my house number, I can call my iPhone 5 right away and it would go through as it should.
Which iPhone do you have? Maybe there's a glitch some where.
