Can't turn on my MacBook Pro...

When I try to turn on my MacBook Pro it stays on the grey screen with the Apple logo and spinning wheel (not the rainbow wheen of death) and hasn't gone past that. Then after a few mintues the Apply logo turns in to a circle with a line through it. I assume this is no0t good news at all. What can I do to fix this?
