Is the backup drive at least twice the data capacity of the original?
I'm having the ame problem. It happens randomly. What does the size of the back up drive being at least twice the size of the original have to do with it?
You should start by using Disk Utility to Repair both souce & destination...
I did that but it did not help.
It may be runing out of space until it deletes older backups for one thing. 🙂

355GB available. Space is not the problem.
How much free space is reported on the backup drive?

Open Console in Applications>Utilities & see if there are any clues or repeating messages when this happens.
For the past month, I get this message every few days: "Time machine couldn't complete the backup to external drive. Uable to complete backup. An error occurred while creating the backup folder."
After deleting the file /Library/Preferences/com.apple.TimeMachine.plist, Time Machine backup works for a day or two - then the cycle repeats itself alll over again, "..couldn't complete backup...".
Disk capacity is 500 GB with 150 GB available.
When I try to verify & repair external drive, error message says: "Volume repair complete. Updating boot support partitions for the volume as requirted. ERROR: Couldn't unmount disk."
Is Spotlight trying to index that drive?
How do I check that?
System Preferences>Spotlight>Privacy pane... add the drive there.
No, spotlight is not indexing the drive.
I have erased my external drive (it had 150/500 MB available) and since then completed two successful backups.
We'll see.
