One of the USB ports on my Macbook Pro isn't working. Any way I can fix this or will I have to take it into the store to get repaired?

Reset the SMC and PRAM

SMC Reset:

1. Shut down the MacBook Pro.
2. Plug in the MagSafe power adapter to a power source, connecting it to the Mac if its not already connected.
3. On the built-in keyboard, press the (left side) Shift-Control-Option keys and the power button at the same time.
4. Release all the keys and the power button at the same time.
5. Press the power button to turn on the computer.

PRAM:

1. Shut down the MacBook Pro.
2. Locate the following keys on the keyboard: Command, Option, P, and R.
3. Turn on the computer.
4. Press and hold the Command-Option-P-R keys. You must press this key combination before the gray screen appears.
5. Hold the keys down until the computer restarts and you hear the startup sound for the second time.
6. Release the keys.

will this delete any of my files?

No.

If that doesn't work, you can unplug all of your USB cables, reboot, and insert the cables again. This resets the USB mapping.

You should get the same results by resetting PRAM.

This (PRAM & SMC reset) works for the front USB port but not the rear - any other suggestions?

If you've updated the Macbook with the firmware update, and done both PRAM/SMC... and still no result... My friend, the port is fried, take it into the genius bar..
