As a test, shut down (power off) your computer and disconnect all USB devices, including hub, except for standard keyboard and mouse (if normally used). If your computer is a laptop, connect its power adapter (so that it's not running on battery power). Do this to reset the shuffle

Start up computer, run iTunes, and connect shuffle to a direct USB port on the computer.
If the replacement USB cable is a third-party product (not the official Apple replacement), make sure it is described as being for the 3rd and 4th gen shuffle. A USB cable that is for the 2nd gen shuffle will not work, because the connector is different.

From the way you answered, I'm not sure if you are answering that question, or saying that the light does not come ON when you move the power switch on the shuffle to the ON position.
Also, what happens when you connect the shuffle to your computer by itself, with nothing else connected to USB?
If you get no response (no light) from the shuffle when you connect it to USB by itself, trying both the original USB cable and a new one that is compatible with the 4th gen shuffle, try connecting something else to the USB port to confirm it is working properly. It would be best to test the USB port with something similar, such as a USB flash drive or external drive. If you can confirm the USB port works properly, it is likely that the cause for this problem is on the shuffle, such as a faulty headphones jack or bad logic board.
If the shuffle is less than one year from purchase date, it is under warranty.
