I found that the noice cancelation microphone on the back of my iPhone 5s is also interfering with my voice, callers can't hear me, but i've noticed that when i cover the microphone, they can hear me fine, so I tried switching off noise cancelation in settings, but this has no effect. Could this be a possible hardware issue, or can a reset fix this issue?

Difficult to say,
Try a reset by hold both buttons till you see the apple logo. If that dont solve the issue try a clean restore according to ht1414. If that dont solve the issue contact applecare for further instructions.
Reset didn't work, i've tested when on a phone call, when i cover the noise canselation mic next to the flash, it work fine, so it's cancelling out my voice essentially, maybe a voice cancellation chip issue, or something, but will see if i can get an exchange, only had the phone a couple of days.
I do have exactly the same problem, however I have investigated it and found out that it works fine while you keep left borrom corner of the phone on your palm so microphone gets reflected sound from your palm it seems
But once I move my palm away from left bottom corner they cant hear me..
I would like to see whether this is a feature of a bug ..
