My Seagate Backup Plus 500 Gb external hard drive runs whenever the computer is in use, but I have set it to back up only once per day at 3:00 a.m.
Does it need a firmware update? Other solution?

Two things to check...
Is that drive added to Spotlight's privacy pane?
In System Preferences>Energy Saver>Sleep tab, is put Hard Drives to sleep when possible checked?
Did both of the recommendations, didn't appear to make a difference as the HD is still running when the iMac is awake.

Might try a script to unmount/mount the drive to see if that helps...
