Q: I purchased Itunes Match and have it on the computer. How to I tell it to turn on now?

I purchased itunes match on my desktop yesterday. I have updated my phone and turned on itunes match in settings. Do I need to turn on or update itunes match through itunes on the computer to have it work?

If so, how do I do that?

iPhone 3G, iOS 4.2
