I'm planning to build a PC containing a GTX 780 Ti, which has a Display Port. I'm planning to use a mini-display port converter and a display port cable to connect it to my iMac 27" (Late 2009). Is this possible? Has anyone attempted this before? Or would I have to use another monitor?

You both need to carefully read Apple's FAQ on Target Display Mode, please click Target Display Mode: Frequently Asked Questions (FAQ) for it.
Just to let you all know that I had a 970 GTX which worked perfectly with the old display port on my late 2009 27" iMac. Though I've since replaced my Mac with a new iMac 5k
iMac 5K's do not support TDM.
I know - just letting people know even the latest nVidia cards work fine with Mac's supporting TDM
