Will I know if someone called or texted me while I had do not disturb on once I've turned it off?
Are those calls/messages just lost or do they just come through after DNS is turned off?

Actually, the calls and messages will arrive on the phone, you just will not be notified. You should find messages in your messages app, and missed calls in your Notification Center, along with any voicemail that might be left.
