What's the best way to use airport express and airport extreme alongside an existing wireless network?

I have a wireless network already in place and I have previously bought Airport Express (to achieve airplay) and Airport Extreme (to apparently boost signal of existing wi-fi and act as an extra router).

Does anyone know the best way to connect all of this up for maximum benefit? I will eventually probably get a time capsule and Apple TV.
