Which hole is that? You have the front/lo-res camera, the ear detector (locks the screen when calling so your cheek doesn't auto-dial) and the ambient light sensor. 3 in all, besides the speaker.
And on the other side you have the 8MP camera, the LED flash and the ambient/noise-cancelling mic.
I mean the small hole on the left side of the earpeace (call speacker). The top is the self camera.
Ist it ok to cover the self camera and that hole? Because yesterday, I just bought a glass screen protector and it is having hole only for the earpeace.
Just make sure the iPhone's screen is perfectly clean and dust free before sticking the protector on. Else your front camera will have a permanent smudge on whatever it photographs. As for the other two (ambient, ear detector), only issue you may have is that the sensitivity gets reduced: the screen doesn't block when you call and put the earpiece to the ear, or auto-dimming is unsatisfactory.
I have a LifeProof frē on mine and that covers up EVERYTHING in order to achieve its 6-foot immersion capability. I've only had to press it a little harder against the ear in order for the screen to block.
Do you mean the microphone (left) or speaker (right) ?
I don't recommend you to do this, if your iPhone will receive damage from this warranty is lost.

The hole left to the earpiece is the ambient light sensor or proximity sensor. I strongly recommend NOT to cover any sensors and/or cameras.
