I have a mid 2011 imac 21.5. My question is, can I run three screens simultaneously, by daisy chaining the one thunderbolt cable?

Yes, if at least one is a Thunderbolt display.
You can't connect a non-Thunderbolt display or adapter directly to the port on the back of a Thunderbolt display. It will work if you connect a Thunderbolt disk drive (with two Thunderbolt ports) between the two displays.
