Purchased items not transferred to library, even after backup.

ITunes continually states “purchased items on the iPad that has not been transferred to library. Should transfer before update iPad.” I initiate ‘backup’ but I still get the same message. How else do I ensure everything has been backed up and that I won’t lose anything.
