All photos and videos lost after first sync on Iphone 5s and cant retrieve after backup

Hi...new to this so please be gentle....plugged in my Iphone 5S to upload holiday photos and videos, the phone has sync'd them? and i clicked on backup and now i cant retrieve them at all?...tried to reset to the backup time but they are gone...any ideas or are they gone for good?

Any help would be useful thanks.

iPhone 5s, iOS 7.1
