It sounds like you are unable to lock the screen with either the Sleep/Wake button, or with the Assistive Touch button on the screen but the Sleep/Wake button will allow you to restart your iPad and will turn it on. I would start by quitting all the running apps on your device:
iOS: Force an app to close

Double-click the Home button.
Swipe left or right until you have located the app you wish to close.
Swipe the app up to close it.
When you have done that and restart the phone and test the issue again:
iOS: Turning off and on (restarting) and resetting

And restore it first as a new device, and verify that it works properly, then restore from your backup with the same article below to ensure that it is still working.

If it does not work correctly when you set up the phone as a new device, I would next seek service for your iPad:
