I have a question about Iphone 5S.
I would like to know: does the Home button have any durability issue ? I read about some people who have using the AssistiveTouch, cause they fear cause any issue just using normally the Home button. Is that true ? Can this button break up just for "using"?

Eventually, of course. Nothing lasts forever. It doesn't make sense not to use something simply because it is certain to wear out one day.
My oldest iPod Touch is nearly six years old. It used the same Home button design as the original iPhone and it still works fine. Since that time, Apple has only improved the Home button design.
Apple does not publish the cycle life limit of the Home button, but it must be on the order of at least tens of thousands.
Of course any mechanical button can eventually break but to use touch ID doesn't require the button to be phisically pushed, you simply place your finger on it so there's no mechanical action used on the button. To not use touch ID for fear of breaking the button is silliness.
As there's no longer an edit button I should clarify that there's no 'additional' mechanical action on the button to use touch ID.
