You do that in Contacts. Mail will then fill in the group when you type the group name in Mail.
Contacts/File/New Group, then select the cards you want and drag them into the group.
Adding to Erics info.... If you hold down the Option key when selecting a contact in Contacts, it will highlight the groups the contact exists.
