Some tests to run when this is happening. From one of the machines that is responsive, attempt to ping the metadata interface of one of the units that is not responding. I will guess that you will not be able to hit the system. If you can not communicate over the metadata network, then this would explain in.
I recall in the 10.6.x days that my MDCs would on occasion stop routing on the metadata network. If I moved the cable to another port in the switch, then it would pick up again. Turned out I had a slowly failing switch. Replacement of the switch resolved the oddities.
Are you using fixed addresses for your MDCs and clients on both the LAN and metadata networks?
