I have been mounting Compact Flash cards on my Mac Pro for many years. Now suddenly they won't mount. Have tried two different readers and different USB slots which I know work. I can mount the card on my MacBook Pro fine so it is possible. Any ideas on how I can get my Mac Pro to mount the cards again?

Does the reader and/or cards show in System Report under USB?
Do they show in Disk Utility?

I managed to get a sporadic mount. Then I couldn't get it to do it again. When not mounted it also doesn't appear in System Report or Disk Utility. Have never done an SMC reset - how much does it zap? Have done many PRAM reset - will try one.

The SMC rest is relatively benign. The lnk discusses it.
Have you tried other USB ports?
Do the USB port(s) work OK for other devices? Maybe yo have a poor connection somewhere

All the USB ports seem to work fine and have been used for other devices.

I'm having the exact same issue today. Worked fine yesterday and many years before. Memory stick has the same problem. Both show up in Disk Utility but the subfolder (the important one) shows as unmounted and won't mount. The memory stick (which has worked fine for years) worked again when I reformatted it from FAT to OS journaled. Can't do that with the CF cards though so not an option. Flummoxed!
I suddenly began having the same problem just this summer on my MBP 15" Retina Late 2013, and none of the resets helped at all. Finally bought a specifically USB 3.0 multireader, rather than the "compatible" one I was using, and it mounted just fine.
