Drag the music there. If you’re moving an iTunes library, launch iTunes with the Option key held down and choose it afterwards.

Note there is a difference between moving just your media folder (which leaves some iTunes files on the internal drive) as one poster recommended and moving your entire library as another poster incompletely covered.  If you are essentially moving all your media off the computer there is no advantage to moving only media and several disadvantages.  In that case move your entire library this way if you use iTunes' default preferences settings:  Copy the entire iTunes folder (and in doing so all its sub-folders and files) intact to the other drive.  Hold down the option (alt) key (shift on Windows) and open iTunes.  At the prompt to create or choose a library choose the copied iTunes folder.  You will need to have that drive turned on each time before starting iTunes or it will revert back to the internal drive and you will need to repeat the option+start.
